What is Thymosin Alpha-1?
Thymosin alpha-1 is a 28 amino acid peptide first isolated from thymic tissue, corresponding to the N-terminal region of prothymosin alpha. Research has examined its reported effects on T-cell maturation and function, dendritic cell activity and cytokine regulation, and it holds pharmaceutical licences in several countries for immunomodulatory indications.

Thymosin alpha-1 (TA1, thymosin α1), INN thymalfasin, is a 28-residue peptide with an acetylated N-terminus. The sequence is Ac-SDAAVDTSSEITTKDLKEKKEVVEEAEN, molecular weight 3108.28 g/mol, CAS 62304-98-7. Goldstein and colleagues (Proc Natl Acad Sci USA, 1977) isolated it from calf thymosin fraction 5 and described a heat-stable, highly acidic peptide. It matches the N-terminal 28 residues of prothymosin alpha.
The sequence has no cysteine or methionine, so there are no disulfides and no methionine oxidation to manage. Unlike most research peptides, TA1 has a pharmaceutical history. As Zadaxin it is a prescription medicine in more than 35 countries, including for chronic hepatitis B. It has never been licensed in the UK, the EU or the United States. Thymosin Alpha-1 Research Applications
- Dendritic cells: in mouse dendritic cells exposed to Aspergillus fumigatus, TA1 drove maturation and IL-12 production through MyD88-dependent Toll-like receptor signalling (Romani et al., Blood, 2004).
- Infection models: in the same study, TA1 protected highly susceptible transplanted mice from aspergillosis.
- Immune markers: Garaci (Ann N Y Acad Sci, 2007) reviews increased MHC class I and Toll-like receptor expression and cytokine production, and combination work with cytokines and chemotherapy.
- Oncology: a 2025 retrospective analysis of 386 patients with platinum-resistant recurrent ovarian cancer studied TA1 alongside PD-1 or PD-L1 inhibitors and chemotherapy (Wang et al., Cancer Manag Res, 2025).

Typical in-vitro work covers dendritic cell maturation, Toll-like receptor signalling and cytokine assays. Much of the older immunology literature is small and mixed, and the recent oncology data are largely retrospective.
Thymosin Alpha-1 vs TB-500
The two share a name and a tissue of origin, and little else. Thymosin alpha-1 is a 28-residue, N-acetylated peptide of 3108.28 g/mol, studied in immunology: dendritic cell signalling and cytokines. TB-500 is the research-market name for synthetic thymosin beta-4, a 43-residue actin-binding protein of 4963.4 g/mol. Its literature concerns actin, cell migration and angiogenesis, built around the seven-residue LKKTETQ actin-binding motif (Philp et al., FASEB J, 2003). Choosing between them means choosing between an immunology tool and a cytoskeletal one.
Research Overview
Goldstein and colleagues (1977) identified TA1 as one of several peptides in thymosin fraction 5. It is strongly acidic, with nine acidic residues against four lysines.
Zadaxin's licence rests on trials in chronic hepatitis B. One randomised trial allocated 98 patients to three groups (Chien et al., Hepatology, 1998), and Camerini and Garaci (Expert Opin Biol Ther, 2015) review the history. No thymosin alpha-1 product is an approved medicine in the UK. Our 5mg vial holds the same molecule for smaller designs.

Storage and Reconstitution
Move vials to the freezer on arrival and keep them dark and dry. Deamidation of the C-terminal asparagine speeds up at higher pH and temperature. Keep the solution cold, limit time at room temperature, keep it away from alkaline solutions, and freeze single-use aliquots for longer storage.
Vials are sold on their own, without bacteriostatic water for mixing or other supplies.
How to reconstitute and handle
Let the vial reach room temperature. Wipe the stoppers of the peptide vial and the bacteriostatic water vial with an alcohol swab. Add the water slowly down the inside wall of the vial, not onto the powder. Swirl gently until the solution is clear and colourless, and do not shake or vortex. The chain ends in asparagine, a residue that can deamidate in solution, so reconstitute promptly and refrigerate. Label the vial with the date.
- No disulfide or methionine handling is needed. Work aseptically with sterile syringes and alcohol-swabbed stoppers.
- Keep reconstituted solutions cold and use single-use aliquots instead of opening one vial repeatedly.
