What is Thymosin Alpha-1?

Thymosin alpha-1 is a 28 amino acid peptide first isolated from thymic tissue, corresponding to the N-terminal region of prothymosin alpha. Research has examined its reported effects on T-cell maturation and function, dendritic cell activity and cytokine regulation, and it holds pharmaceutical licences in several countries for immunomodulatory indications. Thymosin Alpha-1 5mg holds half the lyophilised mass of our 10mg vial, in the same single-vial glass format. The molecule is identical: thymosin alpha-1 (TA1, INN thymalfasin), the 28-residue N-acetylated peptide Ac-SDAAVDTSSEITTKDLKEKKEVVEEAEN, 3108.28 g/mol, CAS 62304-98-7. Goldstein and colleagues (Proc Natl Acad Sci USA, 1977) first isolated it from calf thymus.

The 5mg size suits pilot runs, screening work and designs that need less total peptide. At the same fill volume it gives half the concentration of the 10mg vial.

Thymosin Alpha-1 Research Applications

  • Dendritic cells: in mouse dendritic cells exposed to Aspergillus fumigatus, TA1 drove maturation and IL-12 production through MyD88-dependent Toll-like receptor signalling (Romani et al., Blood, 2004).
  • Immune markers: Garaci (Ann N Y Acad Sci, 2007) reviews increased MHC class I and Toll-like receptor expression and cytokine production.
  • Oncology: recent retrospective work studies TA1 alongside PD-1 or PD-L1 inhibitors and chemotherapy in platinum-resistant recurrent ovarian cancer (Wang et al., Cancer Manag Res, 2025).
Study details

Applications match the 10mg vial: dendritic cell maturation, Toll-like receptor signalling and cytokine assays. Vial size changes logistics, not endpoints. Much of the older immunology literature is small and mixed, and the oncology data are largely retrospective.

Thymosin Alpha-1 5mg vs 10mg

Both vials hold the same molecule, so this is a logistics choice. Each size is its own batch with its own certificate: the 5mg batch is UKPL-9508. At the same fill volume the 5mg vial gives half the concentration, so scale volumes instead of carrying them over. The 10mg vial costs less per milligram and puts twice the work under one batch. The 5mg vial costs less up front, is used up sooner once reconstituted, and leaves less spare material if a design changes. A large study run on 5mg vials spans more batches, so track batch numbers across the dataset.

Research Overview

The molecule is strongly acidic, with nine acidic residues against four lysines, and it has no cysteine or methionine. Goldstein and colleagues (1977) identified it as one of several peptides in calf thymosin fraction 5.

As Zadaxin, thymalfasin is a prescription medicine in more than 35 countries, with indications including chronic hepatitis B. The trials behind that licence include a randomised trial of 98 patients (Chien et al., Hepatology, 1998), and Camerini and Garaci (Expert Opin Biol Ther, 2015) review the history. It has never been licensed in the UK, the EU or the United States.

Storage and Reconstitution

Move vials to the freezer on arrival and keep them dark and dry. The C-terminal asparagine can deamidate in solution, faster at higher pH and temperature. Keep the solution cold, keep sessions short, and freeze single-use aliquots for longer storage.

Vials are sold on their own, without bacteriostatic water for mixing or other supplies.

How to reconstitute and handle

Let the vial reach room temperature. Wipe the stoppers of the peptide vial and the bacteriostatic water vial with an alcohol swab. Add the water slowly down the inside wall of the vial, not onto the powder. Swirl gently until the solution is clear and colourless, and do not shake or vortex. Set the fill volume from your target concentration: 5mg reaches half the concentration of 10mg at the same volume. We give no fill volume and no dosing information, because that is a protocol decision.

  • No disulfide or methionine handling is needed. Work aseptically with sterile syringes and alcohol-swabbed stoppers.
  • Keep reconstituted solutions cold and use single-use aliquots instead of opening one vial repeatedly.
