Full description
CJC-1295 without DAC, also called modified GRF (1-29), is a GHRH (1-29) analogue with four substitutions. It acts at the GHRH receptor on pituitary somatotrophs. Ipamorelin is the pentapeptide Aib-His-D-2-Nal-D-Phe-Lys-NH₂, a selective agonist at the ghrelin receptor, GHS-R1a. The two receptors sit on the same cell but signal differently, which is why the pair is the standard two-receptor design in growth hormone release research.
Equal mass is far from equal moles. At 3367.9 and 711.9 g/mol, 10mg of each is about 3.0 µmol of CJC-1295 against 14.0 µmol of ipamorelin, roughly 4.7 to 1 in favour of ipamorelin.
CJC-1295 + Ipamorelin Research Applications
- Two-receptor design: in 18 men, a GH-releasing peptide plus GHRH gave a greater-than-additive growth hormone response (Bowers et al., 1990).
- Ipamorelin selectivity: in swine it released growth hormone without raising ACTH or cortisol beyond GHRH levels, unlike GHRP-6 and GHRP-2 (Raun et al., Eur J Endocrinol, 1998).

Typical preclinical uses are dual-receptor somatotroph stimulation, pulsatile growth hormone release kinetics with both inputs applied together, and downstream IGF-1 axis work. The fixed mass ratio removes ratio-preparation error, and one batch covers a whole replicate series.

Research Overview
The CJC-1295 in this vial is the no-DAC peptide, often called tetrasubstituted or modified GRF (1-29). It carries D-Ala at position 2, Gln at 8, Ala at 15 and Leu at 27. D-Ala2 blocks cleavage by dipeptidyl peptidase-4, the enzyme that inactivates native GHRH. It has no albumin-binding linker, so it is not the long-acting molecule studied by Teichman and colleagues (J Clin Endocrinol Metab, 2006). Check which form a paper used before applying its findings.
Raun and colleagues (Eur J Endocrinol, 1998) found ipamorelin within a series that lacks the central Ala-Trp of GHRP-1. In rat pituitary cells it released growth hormone with potency and efficacy close to GHRP-6. In swine it left FSH, LH, prolactin and TSH unchanged. It did not raise ACTH or cortisol beyond GHRH levels, even at more than 200 times its ED50.
The case for pairing comes from Bowers and colleagues (1990). In normal men, a growth hormone releasing peptide and GHRH together gave a greater-than-additive growth hormone response, which suggests the two act independently. No controlled study of this specific blend has been published.

Storage and Reconstitution
Keep sealed vials at -20°C, away from light and moisture. After reconstitution, keep the solution at 2-8°C and use it within 4 weeks, or freeze single-use aliquots at -20°C. The GHRH analogue is the less stable partner and sets the in-use window for the whole vial, so aliquot at reconstitution.
Vials are sold on their own, without bacteriostatic water for mixing or other supplies.
How to reconstitute and handle
Treat the vial as one 20mg preparation, with the fill volume set by the concentration your protocol needs. Let the vial and the bacteriostatic water reach room temperature and swab both stoppers. Add the water slowly down the inside wall and swirl gently until dissolved. Do not shake or vortex: the GHRH-class peptide is the shear-sensitive partner. The solution should be clear and colourless. The peptides cannot be separated afterwards. Label the vial with fill volume, concentration and date.
- Work under aseptic conditions with sterile syringes and alcohol-swabbed stoppers. Split reconstituted stock into single-use aliquots and avoid repeated freeze-thaw cycles.
